Determination of glucosylalbumin by phenol-sulphuric acid reaction in hyperglycaemia

Abstract

glucosylalbumin levels in sera of normal and diabetic patients were determined by the phenol-sulphuric acid colorimetric method after separation of albumin by chromatography on Blue-Sepharose. Glucosylalbumin level in diabetics (4.43±1.34 moles/mole) was significantly elevated as compared to control subjects (1.29±0.49, P<0.001). Mean increase in glucosylalbumin (245%) was more than the mean increase of glucosylhaemoglobin (162%) in diabetics. The concentration of glucosylalbumin correlated well (r=0.88, n=49) with glucosylhaemoglobin values. This method for the determination of glucosylalbumin was more sensitive and less time consuming than the thiobarbituric acid method. Significant fall in the glucosylalbumin levels on the fourth day after initiation of therapy in diabetics indicated that this parameter could be a useful index in monitoring regulation of hyperglycaemia.
