Experimentally induced arsenate toxicity attenuated by ixora coccinea flower extract in IEC-6 cells

Abstract

Chronic exposure to arsenic in drinking water has put many at risk of cancer and non-cancerdiseases. In the current study, IEC-6 cells were used as mode to understand the cytotoxicity of arsenate. The aspects of cell survival, intracellular ROS, mitochondrial membrane potential, DNA damage and apoptosis were employed to understand the cytotoxicity. Ixora coccinea is commonly grown in the regions of India. It belongs to the family Rubiaceae. The roots, leaves, stem and the flowers have therapeutic value and used in treating ailments in the traditional system of medicine, folk medicines and in Ayurveda. Rutin is an active component in water extracts and is a well-known antioxidant. Pharmacological studies indicate that the Ixora coccinea has antioxidant, antibacterial, gastroprotective, antidiarrhoeal and hepatoprotective properties. HPTLC chromatographic profile indicated the presence of rutin. Incubation of IEC-6 cells with sodium arsenate (Na2AsO4) induced intracellular ROS production, altered MMP and caused single strand breaks of DNA. Exposure of sodium arsenate also increased apoptosis. Pretreatment of IEC-6 cells with Ixora coccinea flower aqueous extract along with arsenate ameliorated the altered MMP, cell viability, level of intracellular ROS, and DNA damage. Antioxidant enzymes, SOD and catalase was restored indicating the ameliorating behavior of Ixora coccinea aqueous extract.
